By Car

From Manhattan, North Brooklyn, North Queens & the Bronx:

From the Bronx, take the Throggs Neck Bridge to Queens. Stay on I-295 until you come to the L.I.E. Get off and go east on the L.I.E. in the direction of Riverhead.

From North Brooklyn, you should take the I-278/Brooklyn Queens Expressway (B.Q.E) to I-495/Long Island Expressway (L.I.E.) and go east on the L.I.E. in the direction of Riverhead.

From Manhattan, take the Queens Midtown Tunnel. This will turn into the L.I.E. Continue on the L.I.E. eastbound in the direction of Riverhead.

Once you are on the L.I.E., take it east to Route 135/Syosset-Oysterbay Expressway in the direction of Syosset (North). Get off at exit 14E.

Travel down Route 25/Jericho Turnpike (under 5 minutes) until you get to South Woods Road. Take a left onto South Woods Road & drive until you see the entrance for the golf course on your left (right after you pass Shannon road on the right).

From South Brooklyn & Staten Island:

From Staten Island, take the Verrazano bridge to the  Belt Parkway (East). 

Take the Belt Parkway (East) to the Southern State. Take the Southern State to Route 135/Syosset-Oysterbay Expressway in the direction of Syosset (North). Get off at exit 14E.
Travel down Route 25/Jericho Turnpike (under 5 minutes) until you get to South Woods Road. Take a left onto South Woods Road & drive until you see the entrance for the golf course on your left (right after you pass Shannon road on the right).

By Public Transport

From JFK:
It's simple. :)
Take the AirTrain to Jamaica Station, where you'll transfer to the Long Island RailRoad (L.I.R.R.). See below for directions from the LIRR.

From LaGuardia:
It's complicated. :( 
The least stressful way is to take a NYC Yellow Cab to the L.I.R.R. (Jamaica Station). Directions from the LIRR are included below.

L.I.R.R. (from Jamaica Station):

At Jamaica Station, buy tickets to Syosset (pronounced "Si-o-set") at the MTA kiosks. Syosset is on the Port Jefferson Line - which you need to know when checking for your platform on the board. The platform number for the train will most likely be at 4 or higher.

L.I.R.R. (from the city)

The best station to get the train from is Penn Station (34th street at 8th Ave or 7th Ave) in Manhattan. You can get the train direct to Syosset from there. Leave plenty of time before your train as the station is large and can be a bit tricksy to find your way.

You can also get the LIRR at Atlantic Terminal in Brooklyn. However, you will need to change trains at Jamaica Station to get to Syosset. It's pretty easy, but obviously adds to time a little.

Once You Arrive to Syosset

Your best option is to use a car service to the venue or your hotel. Depending on the time of day, there may be taxis at the station at a taxi rank.
